ADAMTS-13 cleaves a domain of ultra-large vWF multimers. Defects in activity or antigen levels of ADAMTS-13 are thought to be the main cause of the thrombotic disorder thrombotic thrombocytopenic purpura (TTP). Acquired TTP is most frequently caused by auto-antibodies directed against ADAMTS-13, leading to strongly reduced protease activity.
The ADAMTS-13 family of assays from Technoclone GmbH provides researchers with easy tools to identify patients with acute idiopathic TTP, and enables close monitoring of how ADAMTS-13 activity changes in the course of the disease and in response to treatment.
Technozym ADAMTS-13 allows the measurement of ADAMTS-13 activity and antigen from the same sample, suitable for fast and routine measurements. Activity of immobilized ADAMTS-13 is measured by cleavage of a fluorogenic substrate. After washing, remaining ADAMTS-13 antigen is detected with a HRP-conjugated antibody, also using a fluorescent substrate.
Technozym ADAMTS-13 INH is a sandwich ELISA using rADAMTS-13 to capture anti-ADAMTS-13 IgG antibodies present in plasma or serum. Its simplicity, short assay time, and good reproducibility make this assay a solution to previously time-consuming methods for measuring ADAMTS-13 auto-antibodies.
