Technozym TAM Activity Assay is a chromogenic assay for the specific determination of thrombin complexed to alpha2 macroglobulin. alpha2 macroglobulin is a broad protease inhibitor that can trap coagulation proteases within a molecular cage. Accurate analysis of the thrombin-alpha2 macroglobulin concentration is particularly important in thrombin generation experiments especially when different types of anticoagulants are tested and in pediatric blood diseases due to the varying amount of plasma alpha2 macroglobulin with respect to age.

Samples containing thrombin-alpha2 macroglobulin are incubated on 96-well plates coated with polyclonal anti-human A2M antibody. Unbound material is removed and wells washed. Detection of thrombin within the immunoabsorbed alpha2 macroglobulin complex is achieved by incubation with a selective thrombin substrate in the presence of antithrombin + heparin (to neutralize non-specifically bound thrombin). Thrombin-alpha2 macroglobulin enzyme activity is quantified by absorbance at 405 nm.
