Intended Use

Protein S Normal Control Plasma is used for day-to-day control of Protein S assays.

Principle

Protein S is a vitamin K-dependent plasma protein which serves as a cofactor for the anticoagulant activity of activated Protein C in the inactivation of factors V and VIII. Protein S exists in human plasma in two forms, both free and complexed to C4b-binding protein (C4b-BP). Free Protein S may be separated from bound Protein S by a simple precipitation of the complex with polyethylene glycol (PEG). Only the free form of the molecule is known to express biological activity.

Deficiency in Protein S has been associated with thrombosis. In some clinical cases of Protein S deficiency, the deficiency has been one of the free Protein S and not of the total Protein S.

Protein S Normal Control Plasma is a freeze-dried preparation of normal human plasma for which the Protein S antigen level (total and free) and clottable Protein S activity has been determined. The antigen level is assayed by ELISA. The clottable Protein S activity can be measured by using a clotting assay.
