Properties
Antithrombin III (antithrombin) is a major coagulation inhibitor and is identical with heparin cofactor I, a plasma protein needed by heparin to act as an anticoagulant. Antithrombin inactivates thrombin and the activated forms of factors IX, X, XI, and XII, i.e. all the coagulation enzymes except factor VIIa and XIIIa.
Antithrombin is a glycoprotein with a relative molecular mass of 58,000 made up of 432 amino acids in a single polypeptide chain cross-linked by three disulfide bridges. Antithrombin III is produced from human plasma and purified by affinity chromatography on heparin-Sepharose gel. Each unit of plasma has been tested and found to be non-reactive for hepatitis C virus (HCV) and for antibodies to human immunodeficiency virus (HIV-1/HIV-2). To further reduce the risk of viral transmission, a chemical virus inactivation step with solvent/detergent (TNBP/Triton) and heat treatment in solution at 60°C for 10 hours have been included in the production process.
