Composition and purity
Lyophilised powder of human plasmin prepared by activation of plasminogen with a matrix bound activator. Contains phosphate buffer salts, lysine and e-aminocaproic acid.

Specific activity
24-40 CU/vial (batch specific insert enclosed).

Application
Fibrinolytic research reagent, including determination of plasmin inhibitor.

Properties
Human plasminogen, the starting material, is prepared from Cohn supernatant I by affinity chromatography (modified procedure according to Deutsch D & Mertz E.T., Science 170, 1095-1096 (1970)). The plasminogen which has a specific activity of about 15 CU/mg protein is activated to plasmin using a matrix bound activator.

Composition

  • Human Plasmin, 2 mg ± 0.5
  • Phosphate buffer, 0.1 mol/l, pH 7.4
  • Lysine, 0.01 mol/l
  • e-amino caproic acid, 0.4 mmol/l

Specific Activity
CU/mg (CU= casein units according to Sqouris et al. Vox Sang 5,357 (1960)).

Storage and Stability
The lyophilized powder is stable at 2-8°C until the expiry date printed on the label. After reconstitution the solution is stable for 8 hours at 2-8°C, 6 months at -20°C. Refreezing should be avoided.

Enzyme Activity Determination
Plasmin activity can be measured using the following chromogenic substrates:

  • S-2251, 25 mg vial S820332 (820332)
  • S-2403, 25 mg vial S822254 (822254)
