Biopool® Stabilyte Tubes are intended for collection of blood samples for determination of tissue plasminogen activator (tPA) and plasminogen activator inhibitor (PAI-1) activities. Biopool® Stabilyte Tubes is specially recommended for use with the Biopool kits Chromolize® tPA (cat# K1103), for tPA activity and specially recommended for use with Spectrolyse® (pL PAI-1), for PAI 1 activity (cat# K101201). Biopool® Stabilyte Tubes have also been successfully used for determination of other analytes such as tPA antigen, PAI-1 antigen and fibrinogen.

Principle

Tissue plasminogen activator activity is unstable in blood plasma. The activity half life ranges from 1 minutes to 4 hours depending on the level of PAI-1 activity. With other collection methods, i.e. anti-coagulants tri-sodium citrate or EDTA, it is impossible to prevent the fast inhibition of tPA by PAI-1. This results in underestimation of circulatinglevels of tPA activity. Biopool® Stabilyte Tubes contain citrate anticoagulant at low pH (patented). Biopool® Stabilyte Tubes immediately reduce the blood pH to about 5.9, inhibiting PAI 1/tPA complex formation and preserving the components of the fibrinolytic system. The components regain full activity after plasma neutralization.
